Elisa實(shí)驗(yàn)常用方法： 
雙抗體夾心法測(cè)定抗原 

將抗原免疫*種動(dòng)物獲得的抗體吸附于固相表面;

加抗原，形成抗原-抗體復(fù)合物; 

加抗原免疫第二種動(dòng)物獲得的抗體，形成抗體抗原抗體復(fù)合物;

加酶標(biāo)抗抗體(第二種動(dòng)物抗體的抗體); 

加底物。底物的降解量＝抗原量。

用于檢測(cè)未知抗原的雙抗體夾心法: 
1. 包被：用0.05M PH9.牰碳酸鹽包被緩沖液將抗體稀釋至蛋白質(zhì)含量為1～10μg／ml。在每個(gè)聚苯乙烯板的反應(yīng)孔中加0.1ml，4℃過(guò)夜。次日，棄去孔內(nèi)溶液，用洗滌緩沖液洗3次，每次3分鐘。(簡(jiǎn)稱(chēng)洗滌，下同)。 
2. 加樣：加一定稀釋的待檢樣品0.1ml于上述已包被之反應(yīng)孔中，置37℃孵育1小時(shí)。然后洗滌。(同時(shí)做空白孔，陰性對(duì)照孔及陽(yáng)性對(duì)照孔)。 
3. 加酶標(biāo)抗體：于各反應(yīng)孔中，加入新鮮稀釋的酶標(biāo)抗體(經(jīng)滴定后的稀釋度)0.1ml。37℃孵育0.5～1小時(shí)，洗滌。 
4. 加底物液顯色：于各反應(yīng)孔中加入臨時(shí)配制的TMB底物溶液0.1ml，37℃10～30分鐘。 
5. 終止反應(yīng)：于各反應(yīng)孔中加入2M硫酸0.05ml。 
6. 結(jié)果判定：可于白色背景上，直接用肉眼觀察結(jié)果：反應(yīng)孔內(nèi)顏色越深，陽(yáng)性程度越強(qiáng)，陰性反應(yīng)為無(wú)色或極淺，依據(jù)所呈顏色的深淺，以“+"、“-"號(hào)表示。也可測(cè)O·D值：在ELISA檢測(cè)儀上，于450nm(若以ABTS顯色，則410nm)處，以空白對(duì)照孔調(diào)零后測(cè)各孔O·D值，若大于規(guī)定的陰性對(duì)照OD值的2.1倍，即為陽(yáng)性。
