ELISA的基礎(chǔ)是抗原或抗體的固相化及抗原或抗體的酶標(biāo)記。結(jié)合在固相載體表面的抗原或抗體仍然保持其免疫學(xué)活性，酶標(biāo)記的抗原或抗體既保留其免疫學(xué)活性，又保留酶的活性。在測(cè)定時(shí)，受檢標(biāo)本（測(cè)定其中的抗體或抗原）與固相載體表面的抗原或抗體起反應(yīng)。用洗滌的方法使固相載體上形成的抗原抗體復(fù)合物與液體中的其他物質(zhì)分開(kāi)。再加入酶標(biāo)記的抗原或抗體，也通過(guò)反應(yīng)而結(jié)合在固相載體上。此時(shí)固相上的酶量與標(biāo)本中受檢物質(zhì)的量呈一定的比例。加入酶反應(yīng)的底物后，底物被酶催化成為有色產(chǎn)物，產(chǎn)物的量與標(biāo)本中受檢物質(zhì)的量直接相關(guān)，所以可根據(jù)呈色的深淺進(jìn)行定性或定量分析。由于酶的催化效率很高，間接地放大了免疫反應(yīng)的結(jié)果，使測(cè)定方法達(dá)到很高的敏感度。

ELISA實(shí)驗(yàn)操作注意事項(xiàng)：
大部分實(shí)驗(yàn)室在使用酶標(biāo)儀之前是通過(guò)目測(cè)判斷結(jié)果，操作過(guò)程隨意性較大，在使用酶標(biāo)儀后操作必須規(guī)范：
A. 使用移液器加液，移液槍頭不能混用。
B. 洗板要干凈，避免交叉污染。
C. 嚴(yán)格按照試劑盒的說(shuō)明書(shū)操作，反應(yīng)時(shí)間準(zhǔn)確。
D. 請(qǐng)勿將樣品或試劑灑到儀器表面或內(nèi)部，操作完成注意做好清潔工作。
E. 不要在測(cè)量過(guò)程中關(guān)閉電源。
F. 對(duì)于因試劑盒問(wèn)題造成的測(cè)量結(jié)果的偏差，應(yīng)根據(jù)實(shí)際情況及時(shí)修改參數(shù)，以達(dá)到*效果。
G. 使用后蓋好防塵罩。
H. 出現(xiàn)技術(shù)故障時(shí)應(yīng)及時(shí)與廠家，切勿擅自拆卸酶標(biāo)儀。
