elisa實(shí)驗(yàn)?zāi)康?br>     酶聯(lián)免疫吸附測定（enzyme-linked immunosorbent assay 簡稱ELISA）是在免疫酶技術(shù)（immunoenzymatic techniques）的基礎(chǔ)上發(fā)展起來的一種新型的免疫測定技術(shù),ELISA過程包括抗原（抗體）吸附在固相載體上稱為包被，加待測抗體（抗原）, 再加相應(yīng)酶標(biāo)記抗體（抗原）,生成抗原（抗體）--待測抗體（抗原）--酶標(biāo)記抗體的復(fù)合物，再與該酶的底物反應(yīng)生成有色產(chǎn)物。借助分光光度計的光吸收計算抗體（抗原）的量。待測抗體（抗原）的定量與有色產(chǎn)生成正比。

elisa實(shí)驗(yàn)原理
    用于免疫酶技術(shù)的酶有很多，如過氧化物酶，堿性磷酸酯酶，β－Ｄ－半乳糖苷酶，葡萄糖氧化酶，碳酸酐酶，乙酰*酯酶，6－磷酸葡萄糖脫氧酶等。常用于ELISA法的酶有辣根過氧化物酶，堿性磷酸酯酶等，其中尤以辣根過氧化物酶為多。由于酶摧化的是氧化還原反應(yīng)，在呈色后須立刻測定，否則空氣中的氧化作用使顏色加深，無法準(zhǔn)確地定量。
